Influence of sleep disruption on protein accumulation in neurodegenerative diseases

چکیده
Abnormal accumulation of disease proteins in the central nervous system is a neuropathological feature neurodegenerative disorders. Recently, growing body evidence has supported role disruption sleep-wake cycle development, pathological changes and abnormal protein diseases, especially Alzheimer’s Parkinson’s disease. Sleep deprivation promotes proteins. Interestingly, amyloid-? (A?) daily oscillations human cerebral spinal fluid (CSF) cleared more sleep. Both circadian genes hormones are associated with deposition. glymphatic pathway meningeal lymphatics have been shown to play critical A? clearance, which mediated by aquaporin (AQP-4) water channel on astrocytes. The rate clearance different during sleep/wake cycle. Most importantly, rhythms facilitate solutes CSF interstitial an AQP-4-dependent manner, further provides for involvement clearance.

Neurodegenerative diseases contain Multiple Sclerosis (MS), Alzheimer’s disease (AD) and Parkinson’s disease (PD), are characterized by neuronal death and neuronal degeneration in specific regions of the central nervous system (CNS). Microglia are the basic immune brain cells and play a role in homeostasis after inflammation challenge. Microglia involves in Neurodegenerative disease...

Inflammatory processes have been shown to be involved in development and progression of neurodegenerative diseases. Mammalian target of rapamycin (mTOR) involves in various cellular processes including autophagy, apoptosis and energy metabolism. Recently, studies have been shown an association between mTOR pathway and inflammation, supporting the role of the pathway in the pathogenesis of infla...

Circadian clock disruption in neurodegenerative diseases: cause and effect?
Disturbance of the circadian system, manifested as disrupted daily rhythms of physiologic parameters such as sleep, activity, and hormone secretion, has long been observed as a symptom of several neurodegenerative diseases, including Alzheimer disease. Circadian abnormalities have generally been considered consequences of the neurodegeneration. Recent evidence suggests, however, that circadian ...
